Chromatograficzne i spektroskopowe badania utleniania 5-O- i
6-O-glukozydów betanidyny wyizolowanych z surowców roślinnych
W ramach pracy doktorskiej przeprowadzono badania nad termicznym rozkładem barwników gomfreninowych zawartych w soku z owoców B. alba oraz barwników betaninowych występujących w ekstrakcie B. vulgaris, a także zidentyfikowano powstałe produkty termicznej dekarboksylacji i dehydrogenacji. W celu dodatkowej identyfikacji powstałych związków badania przeprowadzono również na wyizolowa-nych, oczyszczonych barwnikach gomfreninowych, a otrzymane wyniki odniesiono do danych uzyskanych dla znanych związków betaninowych. Zidentyfikowano również nowe dekarboksylowane pochodne zawarte w ekstrakcie B. vulgaris. W niniejszej pracy przedstawiono także wyniki kompleksowych badań nad alternatywnym mechanizmem utleniania barwników betacyjaninowych kationami Cu2+ i kationorodnikami ABTS+. Zaproponowano szlaki reakcji utleniania barwników betaninowych oraz zidentyfikowano produkty pośrednie oraz końcowe reakcji na podstawie analiz LC-DAD-ESI-MS/MS, LC-MS-IT-TOF oraz NMR. W celu określenia struktur dekarboksylowanych i dehydrogenowanych pochodnych betacyjanin metodą NMR, opracowano metodę semi-syntezy oraz preparatywnego oczyszczania ksanneobetacyjanin. Zaproponowano również mechanizm reakcji nieenzymatycznego utleniania gomfreniny sugerując, że produktem pośrednim utleniania jest pochodna dopachromowa, a nie - jak w przypadku betaniny - metyd chinonowy. Przeprowadzono także badania nad możliwością tworzenia adduktów pomiędzy sulfhydrylowymi wymiataczami wolnych rodników, takimi jak: glutation, cysteina, cysteamina, N- acetylocysteina i ditiotreitol oraz chinoidami, będącymi reaktywnymi produktami pośrednimi podczas reakcji utleniania betacyjanin (betanidyny, gomfreniny i betaniny).
Chromatographic and spectroscopic studies on the oxidation of betanidin
5-O- and 6-5-O-glukosides isolated from plant sources
The studies presented in the dissertation were performed on isolated and purified betalain pigments present in fruits of Malabar spinach (Basella alba L.) and red beetroot extracts (Beta vulgaris ssp. vulgaris), as well as on oxidized betalain derivatives (xan- and neo-derivatives). The primary goal was to conduct research on single, purified betalain pigments, especially for decarboxylated and dehydrogenated derivatives. Studies on thermal decomposition of gomphrenin pigments present in B. alba fruit juice and betanins present in B. vulgaris extract were performed. The resulting products of thermal decarboxylation and dehydrogenation were identified. In order to confirm the identity of the compounds formed during juice heating, the tests were also carried out on isolated and purified gomphrenin pigments, and the results were compared with the data obtained for standard betanin compounds. New decarboxylated derivatives present in red beetroot extract (RBE) were also detected. A comprehensive study on an alternative mechanism of betacyanin oxidation with Cu2+ cations and ABTS + cation radicals was performed. The oxidation pathways for betanin were proposed and intermediate and final products were identified by LC-DAD-ESI-MS/MS, LC-MS-IT-TOF, and NMR analyses. In order to determine decarboxylated and dehydrogenated betacyanin derivatives structures by NMR method, the semi-synthesis and preparative purification of xanneobetacyanins were developed. The mechanism of non-enzymatic gomphrenin oxidation was also proposed, suggesting that the intermediate during gomphrenin oxidation is a dopachromic derivative, not a quinone methide, as in the case of betanin. The possibility of adduct formation between sulfhydrylic free radical scavengers such as glutathione, cysteine, cysteamine, N- acetylcysteine and dithiotreitol and quinoid intermediates resulting from betalain oxidation (betanidin, gomphrenin and betanin) was also examined.
