Detekcja zjawiska chemiluminescencji za pomocą krzemowych
fotopowielaczy w systemach stacjonarnych mikroprzepływowych
Autor, w swoich badaniach, udowodnił możliwość pomiaru zjawiska chemiluminescencji w systemach stacjonarnych i mikroprzepływowych za pomocą krzemowych fotopowielaczy (SiPM). Zaproponował i porównał różne metody detekcji.
Praca doktorska składa się z trzech głównych części. Pierwsza Przedstawia problemy w pomiarach chemiluminescencji, opisuje proponowane metody pomiarowe i prezentuje prototypowy układ odczytowy. Druga część opisuje układ scalony Application Specific Integrated Circuit (ASIC). Trzecia prezentuje wyniki i analizę pomiarów zjawiska chemiluminescencji w systemach stacjonarnych i mikroprzepływowych.
Prototypowy układ elektroniczny składa się z przedwzmacniacza, układu kształtującego z komparatorem i konwertera ładunku na czas (QTC). System odczytowy pozwala na pomiar amplitudy, ładunku, długości trwania impulsu oraz pozwala na zliczanie zdarzeń.
Sposób działania metod pomiarowych został zweryfikowany za pomocą pomiarów chemiluminescencj luminolu. Badania udowodniły skuteczność zaproponowanych metod. Układ elektroniczny generuje niewiele nakładających się sygnałów i jest dokładny. Zależność między natężeniem światła generowanego przez luminol a jego stężeniem jest liniowa dla każdej metody i ma wysoki współczynnik R2.
Metody zostały porównane na podstawie limitu detekcji oraz czułości. SiPM może zostać użyty w budowie przenośnych urządzeń o mały poborze mocy, gdzie jest wymagana detekcja natężenia światła na poziomie pojedynczych fotonów. Krzemowy fotopowielacz może być integrowany w urządzeniach Lab-on-a-chip lub Micro Total Analysis Systems.
Chemiluminescence Detection Using Silicon Photomultiplier in Stationary
and Microfluidic Systems
The author proved that the Silicon Photomultiplier (SiPM) can be used in the detection of the chemiluminescence in the stationary and microfluidic system. The thesis consists of three essential sections. The first one describes difficulties in the chemiluminescence detection, proposes the detection methods and presents the front-end prototype circuit. The second one describes the Application Specific Integrated Circuit (ASIC) design. The third one discusses the results of the chemiluminescence detection in the stationary and microfluidic system.
The designed front-end prototype circuit consists of a preamplifier, a fast shaper with the comparator and a charge to time converter (QTC). The readout system allows to measure the amplitude, integral, the width of the pulse and to count the events.
The performance of the detection methods was verified in the measurements of the chemiluminescence of the luminol. The research proves that the electronic circuit is able to measure the chemiluminescence. It generates low number of overlapping pulses and is accurate. The relationships between the light intensity and the concentration of the luminol are linear for all methods and have high r-squared parameters. The comparison of the efficiency of the detection methods was performed using the limit of the detection and sensitivity parameter.
The SiPM can be effectively used in small, portable devices with low power consumption in applications where the detection on the level of single photons is required. It can be integrated with the microfluidic systems to build Lab-on-a-chip or Micro Total Analysis
